响应面法优化椴树蜂蜜蛋白提取工艺

中文摘要: 以椴树蜂蜜为原材料,采用响应面法优化椴树蜂蜜蛋白提取工艺。根据中心旋转试验设计,以椴树蜂蜜蛋白含量为指标,采用三氯乙酸沉淀法提取椴树蜂蜜蛋白,选取料液比、提取时间、三氯乙酸浓度进行三因素三水平的响应面试验,并与乙醇沉淀法、硫酸铵沉淀法进行对比。结果显示,3个因素对椴树蜂蜜蛋白提取均有一定的影响,其中三氯乙酸浓度影响最大,料液比和提取时间次之,优化后所得的最佳提取工艺为三氯乙酸浓度17.7%,料液比1∶1.83(g/mL),提取时间37.2 min,椴树蜂蜜蛋白含量最大为99.94 μg/g。
Abstract:Response surface methodology(RSM)was used to optimize the extraction process of tilia tuan honey proteins.According to the design of the central rotation test,the tilia tuan honey protein was extracted by trichloroacetic acid precipitation method by response surface methodology and mono-factorial experiment on 3 factors as follows,ratio of material to liquid,extraction time and the concentration of trichloroacetic acid in 3 levels for each,using the content of tilia tuan honey protein as an indicator,and trichloroacetic acid precipitation method was compared with ethanol precipitation method and ammonium sulfate precipitation method.The results showed that three factors had a certain influence on the extraction of tilia tuan honey protein,among which the concentration of trichloroacetic acid had the greatest influence,followed by the ratio of material to liquid and the extraction time.The optimal extraction process was the concentration of trichloroacetic acid 17.7% under the ratio of material to liquid 1∶1.83 (g/mL),extraction time 37.2 min.The maximum content of tilia tuan honey protein was 99.94 μg/g in that extract environment.
