中文摘要: 通过非标记定量(label-free)蛋白质组学技术对宁夏中宁和青海格尔木两个产地的枸杞干果样品进行蛋白组学比较。结果表明两种样品共鉴别出蛋白质2 477个,中宁产区样品中含蛋白质1 942个(其中174个为中宁样品特有),格尔木产区样品中含蛋白质2 065个(其中297个为格尔木样品特有);另外中宁产地的样品和格尔木产地的样品相比有86个蛋白质的表达量下调,69个蛋白质的表达量上调。对样品蛋白质进行层次聚类分析,证明两产地样品在蛋白质组学上有明显差异。利用GO数据库和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)数据库对所有差异蛋白质进行检索,筛选后得到28个目标差异蛋白质,其中包括:4个金属离子转运蛋白、6个热休克蛋白、3个几丁质酶、4个磷酸肌醇转化酶、2个过氧化氢酶、3个与植物激素调节有关的蛋白质、5个与卟啉和叶绿素代谢相关的酶、1个核糖核苷二磷酸激酶。
Abstract:Label-free technology was used to make a comparison of proteomics between the Ningqi No.1 Lycium barbarum L.dried fruit samples from Zhongning,Ningxia and Golmud,Qinghai.The results showed that a total of 2 477 proteins were identified from the two samples,including 1 942 proteins from Zhongning samples(174 of which were endemic to Zhongning samples),and 2 065 proteins from Golmud samples(297 of which were endemic to Golmud samples).In addition,compared with that of Golmud sample,86 proteins in Zhongning sample were down-regulated and 69 proteins were up-regulated.The hierarchical clustering analysis of the samples showed that there were significant differences in proteomics between the samples from the two producing areas.All the differentially expressed proteins were detected by Gene Ontology(GO)database and KEGG database,and totally 28 target differentially expressed proteins were focused,which include 4 metal ion transporters,6 heat shock proteins,3 chitinases,4 phosphoinositol converting enzymes,2 catalases,3 proteins involved in plant hormone regulation,5 enzymes involved in porphyrin and chlorophyll metabolism,1 nucleoside diphosphate kinase.
