桑树内生真菌的分离、鉴定及其对DNJ生物合成的影响

中文摘要: 从桑树枝叶中分离得到并鉴定内生真菌,对其及合成的1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)进行分析。采用乙醇与升汞漂洗,无菌水冲洗数次的方法对桑树枝叶进行表面灭菌,使用马铃薯葡萄糖琼脂(potato dextrose agar,PDA)培养基分离桑树枝叶中的内生真菌,FMOC-Cl柱前衍生化高效液相色谱法测定DNJ的含量。分离的内生真菌鉴定其与Hypoxylon sp.SUT242具有较高的同源性,DNJ含量的平均值:对照组2.987 μg/mL,试验组3.485 μg/mL,试验组DNJ百分比含量增加16.6%。结果表明,从桑树中分离的内生真菌可能属于Hypoxylon的一个种,能够将桑树枝条中的某些物质代谢转化成DNJ。
Abstract:The endophytic fungi were isolated from mulberry branches and were identified,and the synthetic 1-deoxynojirimycin(DNJ)accompanied by endophytic fungi was analyzed.The surface of mulberry branches was sterilized by washing with ethanol and mercuric chloride,and then by washing with sterile water several times.Endophytic fungi were isolated from mulberry branches by using potato dextrose agar(PDA)medium.The content of DNJ was determined by FMOC-Cl pre column derivatization high performance liquid chromatography.The isolated endophytic fungi were identified to have a high homology with Hypoxylon sp.SUT242;the average content of DNJ in the control group and experimental group were 2.987 μg/mL and 3.485 μg/mL,respectively.The content of DNJ in the experimental group increased 16.6%,which compared to the control group.Endophytic fungi isolated from mulberry may belong to a species of Hypoxylon,could make use of some substances in mulberry branches and transform them into DNJ through metabolism.
