地黄花多糖含量测定及其柱前衍生化HPLC指纹图谱分析

中文摘要: 比较地黄花不同开花时期及盛花后期不同花部位的多糖含量差异;建立地黄花多糖部位柱前衍生化高效液相色谱(high-performance liquid chromatography,HPLC)指纹图谱,明确地黄花多糖的单糖组成,为地黄花的进一步开发提供理论依据。采用苯酚-硫酸比色法测定地黄花多糖的含量;将地黄花多糖水提液盐酸水解后采用1-苯基-3-甲基-5-吡唑啉酮(1-phenyl-3-methyl-5-pyrazolone,PMP)衍生化,建立指纹图谱,分析地黄花不同开花时期及花盛期不同花部位的多糖中单糖组成。结果表明,地黄花花蕾期多糖含量为3.98%~4.93%,盛花期多糖含量为3.65%~3.78%,花蕾期到盛花期多糖含量出现先增加后降低趋势,在花蕾后期出现最大值,且花蕾期平均多糖含量高于盛花期;盛花期花裂片、花筒上部及花筒下部中多糖含量分别为3.84%、4.05%、2.93%,花筒上部平均多糖含量最高。地黄花多糖柱前衍生化高效液相指纹图谱共标定10个共有峰,指认了4个峰,分别为鼠李糖、D-葡萄糖、D-半乳糖和阿拉伯糖,不同开花时期和盛花期不同花部位多糖单糖组成相似,9批地黄花样品相似度均在0.95以上。地黄花花蕾期平均多糖含量高于盛花期,盛花期花筒上部平均多糖含量最高,花裂片其次,花筒下部多糖含量最少。地黄花作为地黄的非药用部位具有一定的药用和食用开发价值。

Abstract:The differences of polysaccharide in different periods and different parts in the late blooming stage of Rehmannia glutionsa flower were compared.The high-performance liquid chromatography(HPLC)fingerprint of the pre-column derivatization of polysaccharides from Rehmannia glutionsa flower was established for the monosaccharide composition of polysaccharides.The research would be provided experimental basis for the further development of Rehmannia glutionsa flower.The content of polysaccharide was determined by phenolsulfuric acid colorimetry.The monosaccharide composition of polysaccharides in different parts of Rehmannia glutionsa flower in blooming stage and in different stages were analyzed by using 1-phenyl-3-methyl-5-pyrazolinone(PMP)derivatization after hydrochloric acid hydrolysis.The polysaccharide content in bud stage was 3.98%-4.93%.The polysaccharide content in blooming stage was 3.65%-3.78%.From bud stage to blooming stage,the content of polysaccharide increased first and then decreased.It reached the maximum at the later stage of flower bud.The average polysaccharide content was higher in bud stage than that in the blooming stage;The contents of polysaccharides in flower lobes,upper part of flower barrel and lower part of flower barrel were 3.84%,4.05% and 2.93%.And the average polysaccharide content of the upper part of flower barrel was the highest.10 common peaks were found and 4 peaks were identified in high-performance liquid chromatography(HPLC)fingerprint of precolumn derivatization,which were rhamnose,D-glucose,D-galactose and arabinose.The monosaccharides in the polysaccharides were similar,and the similarities of 9 samples were all above 0.95.As a non-medicinal part of Rehmannia glutionsa,the medicinal and edible value of its flower should be developed.
