中文摘要: 以鸿雁(Anser cygnoides)骨骼为原料,采用复合酶酶解制备雁骨胶原多肽,在单因素试验的基础上,采用响应面法优化酶解工艺。以水解度为指标,确定木瓜蛋白酶、中性蛋白酶和碱性蛋白酶为最佳酶组成复合酶,质量比为1∶2∶1,最佳酶解工艺为:酶解温度50℃、pH 7.1、加酶量6 100 U/g、酶解时间5 h,此时水解度为26.34%。通过超氧阴离子自由基和羟基自由基的清除能力测定,发现雁骨胶原多肽的抗氧化活性与胶原多肽质量浓度成正相关,当底物浓度为6 mg/mL时,超氧阴离子自由基和羟基自由基的清除率分别为59.21%、51.82%。通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)和分子排阻色谱法测定复合酶酶解制备的雁骨胶原多肽分子量为851 Da~11 092 Da。氨基酸组成分析显示雁骨胶原多肽富含甘氨酸、丙氨酸、脯氨酸、羟脯氨酸及天冬氨酸等,这些特定的氨基酸与胶原多肽的抗氧化活性相关,为雁骨的开发利用提供参考。
Abstract:The bone of Anser cygnoides was used in the experiment that was hydrolyzed by complex enzyme.For preparing the collagen polypeptide of Anser cygnoides bone,the single factor experiment was carried out for determining the optimal enzymatic hydrolysis process combined with the method of response surface.The degree of hydrolysis which was indicated as an indicator expounded that the best complex enzymes formed of papain,neutral protease and alkaline protease with a mass ratio of 1∶2∶1.The optimal enzymatic conditions included temperature as 50℃,pH as 7.1,enzyme amount as 6 100 U/g,time as 5 h,showed the result of hydrolysis degree was 26.34%.For analysed the free radical scavenging ability of Anser cygnoides bone collagen polypeptide,superoxide anion and hydroxyl radical scavenging experiments were carried out,the result suggested that the antioxidant activity of Anser cygnoides bone collagen polypeptide was positively correlated with the concentration of collagen polypeptide.When the substrate concentration was 6 mg/mL,the scavenging rates of superoxide anion and hydroxyl radical were 59.21% and 51.82%,respectively.Sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)experiment and exclusion chromatography showed that the molecular weight of Anser cygnoides bone collagen polypeptide was from 851 Da to 11 092 Da.Amino acid composition analysis showed that the Anser cygnoides bone collagen polypeptide was rich in glycine,alanine,proline,hydroxyproline and aspartic acid and these specific amino acids were related to the antioxidant activity of the collagen polypeptide.
