中文摘要: 设计单因素试验考察海藻酸钠、氯化钙浓度、硬化时间、戊二醛浓度和交联时间对海藻酸钠包埋固定几丁质脱乙酰酶的影响,并通过响应面法优化固定条件。得到最佳固定条件为:海藻酸钠浓度27.8 g/L,氯化钙浓度为31.0 g/L,硬化1.84 h,戊二醛浓度0.025%,交联30 min。对固定化酶的酶学性质进行研究,结果表明相较于游离酶,固定化酶的热稳定性和pH稳定性都有明显提高。固定化酶在循环操作6次后,酶活仍然保持初次酶活性的63.15%,为几丁质脱乙酰酶的工业化应用提供理论基础。
Abstract:Based on single-factor and response surface method experiments,conditions of immobiling chitin deacetylase to sodium alginate were investigated.The optimal conditions of immobilization were obtained as follows:sodium alginate concentration 27.8 g/L,calcium chloride concentration 31.0 g/L,immobilization time 1.84 h,glutaric dialdehyde concentration 0.025%,and cross-linking time 30 min.The enzymatic properties of the enzyme immobilized with optimal conditions were studied.The results showed that compared with the free enzyme,the thermal stability and pH stability of the immobilized enzyme were significantly improved.After the sixth repeat usage,the activity of immobilized enzyme retained 63.15% of the initial value,providing a theoretical basis for the industrial application of chitin deacetylase.
