中文摘要: Box-Behnken响应面法优化超声辅助提取女贞子原花青素工艺。以原花青素的提取率为考察指标,提取时间、超声功率、液料比和乙醇体积分数为考察因素,采用四因素三水平做响应面分析。利用邻苯三酚法、DPPH和水杨酸法对女贞子原花青素抗氧化性进行研究。结果表明,女贞子中的原花青素的最佳提取工艺为:提取时间33 min,超声功率400 W,液料比27∶1(mL/g),乙醇浓度50%。在最佳提取条件下,原花青素的提取率为(2.85±0.06)%。当原花青素浓度为1.0 mg/mL时,对DPPH自由基的清除率可达到97%;对超氧阴离子的清除率可达到60.14%;对羟自由基的清除率可达到78.88%。该研究优化的女贞子原花青素提取工艺合理、稳定,技术可行,提取的原花青素有较好的抗氧化性。

Abstract:To optimize the ultrasonic-assisted extraction technology of proanthocyanidins from the Ligustrum lucidum ait by box-behnken response surface methodology.The single-facotr experiment and response surface methodology were adopted to study the effects of extraction time,ultrasonic power,liquid-solid ratio and ethanol concentration on the extraction yield of proanthocyanidins from Ligustrum lucidum ait.Then the antioxidant activities of proanthocyanidins were evaluated by pyrogallol autoxidation,DPPH and salicylic acid(SA)methods.The results showed that the optimal extraction technology for proanthocyanidins were as following,extraction time was 33 min,ultrasonic power was 400 W,liquid-solid ratio was 27∶1(mL/g),and ethanol concentration was 50%.Under optimal technology,the extraction yield of proanthocyanidins was about(2.85±0.06)%.When the concentration of proanthocyanidins was 1.0 mg/mL,the maximum scavenging rate of DPPH radicals reached to 97%,the maximum clearance rate of superoxide anions reached to 60.14%,and the scavenging rate of flavonoids to hydroxyl radicals was 78.88%.The optimal extraction technology of proanthocyanidins from the Ligustrum lucidum ait was reasonable,stable,feasible and the proanthocyanidins had good antioxidant activity.
