中文摘要: 研究纤维素酶提取肾茶多糖的工艺及体外抗氧化活性。在单因素试验基础上进行响应面法优化提取工艺条件,并通过自由基清除及还原能力试验,测定肾茶多糖的抗氧化活性。结果表明,最佳工艺条件料液比1∶30(g/mL),酶用量2%,酶解pH3.90,酶解温度52℃,酶解时间2 h,肾茶多糖提取率为12.51%。抗氧化活性研究中,肾茶多糖对自由基的清除及还原能力随其浓度的增大而增强。综合分析说明,响应面法优化纤维素酶提取肾茶多糖的工艺有效、可行,所提多糖具有较强的抗氧化活性。
Abstract:The aim of the study was to optimize the cellulase extracting technology of polysaccharides from Clerodendranthus spicatus and its antioxidant activity in vitro.Based on a single factor experiment to carry on the response surface method to optimize the extraction process conditions of polysaccharides.Meanwhile,the antioxidant activity of its was also determined by radical scavenging and reducing power test.The results showed that the optimum extraction condition were material-liquid ratio 1 ∶30 (g/mL),cellulase amount 2%,hydrolysis pH3.90,hydeolysis temperature 52℃ and hydrolysis time 2 h,the yield of polysaccharides was 12.51%.The antioxidant tests showed that the radical scavenging and reducing power of the polysaccharides increased along with the increase of its concentration.Comprehensive analysis indicated that cellusase extracting technology of polysaccharides from Clerodendranthus spicatus by response surface methodology was effective and feasible.The polysaccharides had stronger antioxidant activity.
