中文摘要: 研究白首乌花多糖对酒精性肝损伤小鼠的保护作用,采用超声方法辅助提取白首乌花多糖,并通过体外DPPH自由基清除试验检测其体外抗氧化活性。选取C57小鼠,随机均分为空白组、模型组、阳性药物组、白首乌花多糖低剂量组和高剂量组,空白组小鼠给与常规液体饲料,其余小鼠给与液体酒精饲料10 d,并于第11天上午大剂量高浓度灌胃酒精,9 h后解剖。检测小鼠血清生化指标、肝脏生化指标。结果表明,白首乌花多糖DPPH自由基清除的IC50值为(69.9±0.12)g/mL;相比于模型组,高剂量组可显著地降低小鼠血清谷丙转氨酶(alanine aminotransferase,ALT)39.7%、谷草转氨酶(aspartate aminotransferase,AST)30.3%、总胆固醇(total cholesterol,TC)23.17%、甘油三酯(triglyceride,TG)26.63%、低密度脂蛋白(low density lipoprotein,LDL)21.72%、非酯化脂肪酸(nonesterified fatty acid,NEFA)41.05%(p<0.05);提高小鼠肝脏超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)水平19.967%和 36.72%(p<0.05),降低肿瘤坏死因子(tumor necrosis factor-α,TNF-α),白介素-1β(Interleukin-1β,IL-1β)和丙二醛(malondialdehyde,MDA)含量 32.56%、27.65%和 23.93%(p<0.05)。因此,白首乌花多糖可以有效的缓解小鼠酒精性肝损伤,具有一定的保肝作用,且为相关产品的开发应用提供一定的理论支撑。
Abstract:The protective effect of Cynanchum auriculatum flower polysaccharides on mice with alcoholic liver injury was studied.Ultrasonic method was used to assist the extraction of polysaccharides from Cynanchum auriculatum flower,and its antioxidant activity was detected by DPPH free radical scavenging assay in vitro.C57 mice were randomly divided into blank group,model group,positive drug group,low dose group and high dose group of Cynanchum auriculatum polysaccharide.The blank group mice were given conventional liquid feed,and the other mice were given liquid alcohol diet for 10 days,and in the morning of the 11th day,high doses of high concentration of gastric alcohol,9 h after dissection.Detection of serum biochemical parameters in mice.The results showed that the IC50value of DPPH free radical scavenging of polysaccharides was(69.9±0.12) g/mL.Compared with the model group,the high dose group significantly reduced serum alanine aminotransferase(ALT)39.7%,aspartate aminotransferase(AST)30.3%,total cholesterol(TC)23.17%,triglyceride(TG)26.63%,low density lipoprotein(LDL)21.72%and nonesterified fatty acid(NEFA)41.05%(p<0.05);increased liver superoxide dismutase (SOD)and glutathione peroxidase (GSH-Px)levels in mice 19.967%and 36.72%(p<0.05).The interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and malon-dialdehyde (MDA)contents were decreased by 32.56%,27.65%and 23.93% (p<0.05).Therefore,Cynanchum auriculatum flower polysaccharide could effectively alleviate alcoholic liver injury in mice,had a certain liver-protecting effect,and provided theoretical support for the development and application of related products.
