中文摘要: 利用超声波技术提取青梅中的有机酸,在探究超声温度、超声时间、料液比及超声功率对提取总有机酸得率影响的基础上,用正交试验优化得到最佳工艺;并对比超声波辅助提取液与乙醇回流提取液中,有机酸种类、含量以及香气成分的差异。结果表明,超声波辅助提取最佳条件为:超声温度70℃,超声时间20 min,料液比1∶15(g/mL),超声功率320 W,在此条件下青梅果肉(干重)总有机酸得率为35.07%;采用高效液相色谱(high-performance liquid chromatography,HPLC)测定提取液中8种主要有机酸含量,与乙醇回流法相比,苹果酸、柠檬酸、酒石酸、乳酸和乙酸显著提高;对提取液的香气成分进行气相色谱-质谱(gas chromatography-mass spectrometry,GC/MS)分析表明,青梅有机酸提取液特征香气成分为酸类物质,与乙醇回流相比,超声波提取青梅特征香气成分的提取率和保留率显著提高。
Abstract:Ultrasonic technology was utilized to extract organic acids from Prunus mume.The effects of ultrasonic temperature,ultrasonic time,solid-liquid ratio and ultrasonic power on the extraction rate of organic acids were studied.Based the obtained single factor experiment data and by using orthogonal test method,the extraction process for organic acids was optimized.The difference of organic acids contents and aroma components in the extract solution from ultrasonic assisted extraction and ethanol reflux extraction was compared.The results showed that the optimal ultrasonic assisted extraction conditions were as follows:ultrasonic temperature 70℃,ultrasonic time 20 min,solid-liquid ratio 1∶15(g/mL)and ultrasonic power 320 W.Under these conditions,the maximum extraction rate of 35.07%was obtained.The content of eight main organic acids in the extract was determined by high-performance liquid chromatography(HPLC).Compared with the ethanol reflux extract,malic acid,citric acid,tartaric acid,lactic acid and acetic acid in the ultrasonic assisted extrat were significantly improved.Gas chromatography-mass spectrometry(GC/MS)analysis of the aroma components of the extract indicated that the characteristic aroma components of Prunus mume organic acids were acids.Compared with the ethanol reflux extract,the extraction rate and retention rate of the characteristic aroma compounds in the ultrasonic assisted extrat were significantly increased.
