中文摘要: 该试验以葛根为试验材料,对广西北海、贵港和柳州产的葛根蛋白质的提取工艺及其功能性进行研究。用凯氏定氮法测定葛根总蛋白含量,水提回流法提取葛根蛋白质;建立以牛血清白蛋白为标准品,双缩脲试剂为显色剂,分光光度法测定葛根蛋白质含量的分析方法。结果表明:葛根蛋白质的最优提取条件是提取溶剂pH 8、液料比20 ∶1(mL/g)、提取温度30 ℃和提取时间2.5 h;三地葛根中蛋白质的提取率为广西北海野葛58.87 %、贵港野葛83.20%和柳州粉葛22.27%。葛根蛋白质功能性研究结果表明:葛根蛋白质的持油力4.06 g/g,持水力4.49 g/g,乳化性34.88%,起泡性6.67%。
Abstract:In this experiment,Pueraria lobata was used as the experimental material to study the extraction process and functionality of Pueraria lobata protein produced in Beihai,Guigang and Liuzhio of Guangxi.Guigang and Liuzhou,located in Guangxi,taking Pueraria lobata as experimental material.The total protein content of Pueraria lobata was determined by kjeldahl method and the Pueraria lobata protein was extracted by water reflux method.The analytical method for the determination of Pueraria lobata protein content by spectrophotometry was established by using bovine serum albumin as standard and biuret reagent as chromogenic reagent.The results showed that the optimal extraction conditions of Pueraria lobata protein were extraction solvent pH 8,ratio of liquid to material 20 ∶1 (mL/g),extraction temperature 30 ℃ and extraction time 2.5 h.The extraction rate of protein of the Beihai was 58.87%,the Guigang was 83.20%and the Liuzhou was 22.27 %.The functional study of Pueraria lobata protein showed study on that the oil holding capacity of Pueraria lobata protein was 4.06 g/g,the water holding capacity was 4.49 g/g,the emulsifying property was 34.88%,and the foaming property was 6.67%.
