山药多糖提取工艺的响应面法优化及其功能活性分析

中文摘要: 利用Box-Behnken设计-响应面优化山药多糖的提取工艺,初步评价不同产地山药多糖清除1,1-二苯基-2-三硝基苯肼(DPPH·)能力、羟自由基(OH·)能力和超氧阴离子(O2-·)能力和α-葡萄糖苷酶抑制活性能力。以超声辅助提取温度、提取时间和料液比为自变量,以山药多糖提取率为因变量,采用响应面分析法优化山药多糖超声辅助提取工艺:提取温度66℃,提取时间26 min,液料比22∶1(mL/g),在此条件下,多糖得率为9.34%。不同产地山药多糖对α-葡萄糖苷酶活性具有抑制作用,并随着多糖浓度的提高其抑制率随之提高。抗氧化活性试验表明山药多糖对DPPH·、OH·和O2-·具有较显著清除作用,并呈现一定的浓度依赖性。其中4个产地山药多糖中河南怀山药多糖含量最高,对α-葡萄糖苷酶的抑制作用、对DPPH·和OH·自由基的清除能力均最优,预处理河南为山药道地产区质优效佳的传统认知相符。
Abstract:To optimize the extraction technology for Chinese yam polysaccharides by response surface method,and preliminary researched the DPPH free radical scavenging capacity (DPPH·),hydroxyl radical(OH·),superoxide anion radical (O2-·)and alpha glucosidase inhibitory activity.In order to evaluate the indicators,the effects of ultrasonic extraction temperature,extraction time and ratiliquid-solid ratio on Chinese yam polysaccharide yield were compared by Box-Behnken design-response surface method.The optimal ultrasonic extraction technology were determined as follows:extraction temperature of 66℃,extraction time for 26 min,liquid-to-material ratio of 22 ∶1(mL/g),under which the yield of polysaccharide was 9.34%.Polysaccharides from different habitats have an inhibitory effect on alpha glucosidase activity,and the inhibition rate increases with the increase of polysaccharide concentration.The antioxidant activity test showed that polysaccharide had a significant scavenging effect on DPPH·,OH·and O2-·,and showed a certain concentration dependence.The content of polysaccharides in Henan province was the highest,the inhibitory effects on alpha glucosidase activitie and the scavenging activities of DPPH·and OH·were significant.
