中文摘要: 对东北刺人参(ChiRenShen,CRS)体内外抗氧化及延缓衰老作用进行研究。优化微板法测定DPPH 自由基清除率的条件,考察东北刺人参的体外抗氧化作用,同时以果蝇生物模型和小鼠为考察对象,测定不同浓度的刺人参对果蝇寿命相关行为学指标和力竭游泳实验小鼠丙二醛(malondialdehyde,MDA)含量和超氧化物岐化酶(superoxide dismutase,SOD)活力的影响。体外实验结果显示,微板法测定DPPH 自由基清除率的最佳反应时间为30 min,反应体积为1 ∶1,刺人参水煎液DPPH 自由基清除率的IC50 值为0.829 mg/mL,阳性对照药VC 的IC50 值为0.040 1 mg/mL;动物实验结果显示,东北刺人参可延长果蝇寿命,增加果蝇的子代数量,提高老年蝇的攀爬能力,提高老龄果蝇抗亚急性氧化损伤能力,并且可以降低MDA 含量,提高SOD 活力。东北刺人参具有体内外抗氧化及延缓衰老作用。
Abstract:The anti-oxidation and anti-aging effects of wild Oplopanax elatus Nakai.(CRS)in Changbai Mountain were studied.The conditions of DPPH were determined by microplate method.The anti-oxidation effect of CRS in vitro was investigated.The biological model of drosophila was used as the object of investigation.The effects of different concentrations of acanthopanax ginseng on the longevity related behavioral indexes of drosophila melanogaster and MDA content and SOD activity of exhausted swimming mice were determined.The in vitro results showed that the optimal reaction time for DPPH free radical scavenging by microplate method was 30 min,the reaction volume was 1 ∶1,and the IC50 value of DPPH free radical scavenging rate of CRS was 0.829 mg/mL,the positive control Vitamin C had an IC50 value of 0.040 1 mg/mL.The results of in vivo experiments showed that the ginseng lyophilized powder could prolong the lifespan of Drosophila,increase the number of offspring of Drosophila,improve the climbing ability of the old fly,and improve the anti-acute oxidative damage and water content of the old Drosophila,and it could reduce MDA content and improve SOD activity.Changbai Mountain wild CRS had anti-oxidation and anti-aging effects in vitro and in vivo,and could improve the life-related indicators of fruit flies.
