中文摘要: 采用超声波-微波协同法提取蛹虫草多糖,并研究其对小鼠脾细胞增殖的影响,初步评价其免疫活性。通过单因素和L18(37)正交试验研究了物料粒度、料液比、超声波功率、超声波时间、超声提取次数、提取温度、乙醇与浓缩液之比对蛹虫草多糖提取率的影响。正交试验结果表明,超声波功率、物料粒度对蛹虫草多糖的得率均呈现出显著的影响,进而确定蛹虫草多糖提取最优工艺参数:物料粒度0.150 mm,提取次数为3 次,微波功率400 W,超声波功率300 W,超声波处理时间30 min,提取温度70 ℃,料液比1 ∶40(g/mL),乙醇与浓缩液之比4 ∶1(体积比)。在最佳条件下,可得到多糖提取率为6.28%。小鼠脾细胞增殖试验表明,在一定的剂量内,提取到的蛹虫草多糖能明显促进小鼠脾细胞的增殖,表明蛹虫草多糖具有免疫调节活性。
Abstract:The polysaccharides of Cordyceps militaris were extracted by ultrasonic-microwave synergy method,and the effects on the proliferation of mouse spleen cells were studied.The immunological activity was evaluated.The effects of material size,solid-liquid ratio,ultrasonic power,ultrasonic time,ultrasonic extraction times,extraction temperature,ethanol and concentrate on the extraction rate of Cordyceps militaris polysaccharides were studied by single factor and L18(37)orthogonal test.The results of orthogonal test showed that the ultrasonic power and material particle size had significant effects on the yield of polysaccharides from Cordyceps militaris,and the optimal technological parameters of polysaccharide extraction from Cordyceps militaris were determined:material particle size was 0.150 mm,extraction times was 3 times,microwave power was 400 W,ultrasonic power was 300 W,ultrasonic treatment time was 30 min,extraction temperature was 70 ℃,ratio of material to liquid was 1 ∶40(g/mL),ratio of ethanol to concentrate was 4 ∶1(volume ratio).Under the optimal conditions,the polysaccharide extraction rate was 6.28%.The mouse spleen cell proliferation test showed that the polysaccharide extracted from Cordyceps militaris could promote the proliferation of mouse spleen cells at a certain dose,indicated that the polysaccharide of Cordyceps militaris had immunomodulatory activity.
