超声波辅助法提取百香果壳中黄酮的工艺及其抗氧化性研究

中文摘要: 采用超声波辅助法对百香果果壳中的黄酮进行提取,通过单因素试验和正交试验确定最佳提取参数。经过聚酰胺层析柱分离纯化,以VC 为对照,采用邻苯三酚自氧化法、DPPH 和水杨酸法对黄酮的抗氧化性进行研究。结果表明:超声波辅助法提取的最优条件为60%乙醇为提取溶剂、温度50 ℃、料液比1 ∶30(g/mL)、超声时间40 min,超声功率240 W,此条件下提取的百香果果壳黄酮提取率为8.084 0 mg/g。通过聚酰胺层析柱纯化,以70%乙醇为洗脱剂,得到的样品纯度为20.08%,回收率为82.95%。黄酮对羟基自由基的最大清除率可达58.66%,对DPPH 自由基的最大清除率可达93.81%,对超氧阴离子自由基的最大清除率可达82.61%。试验结果表明百香果果壳黄酮具有较好的抗氧化性,是一种理想的天然抗氧化剂,试验结果为百香果的综合开发提供一定的科学依据。
Abstract:The extraction conditions of ultrasonic method were studied on flavonoids from shells of passion fruit by single factor and orthogonal test.The flavonoids were purified by using column chromatography with polyamide.The antioxidant activity of flavonoids was studied by using the pyrogallol autoxidation method,DPPH and salicylic acid(SA),and vitamin C(VC)in the same concentration was compared as control group.The results showed that the optimal extraction conditions of flavonoids from shells of passion fruit were 60%ethanol as extraction agent,the temperature of 50 ℃,solid-liquid ratio of 1 ∶30(g/mL),ultrasonic extraction time of 40 min and ultrasonic power of 240 W.Under this optimal condition,the flavonoids content was 8.084 0 mg/g.The purity of the sample and the recovery respectively were 20.08 % and 82.95 % with 70 % ethanol as the eluant by using polyamide column chromatography.The scavenging rate of flavonoids to hydroxyl radicals was 58.66%,the maximum scavenging rate of DPPH radicals reached to 93.81%,and the maximum clearance rate of superoxide anions reached to 82.61%.The results showed that flavonoids from shells of passion fruit exhibited high antioxidant activity and were an ideal natural antioxidant.The experimental results could provide some scientific basis for the comprehensive utilization of passion fruit.
