南瓜均一多糖的分离纯化及其抗氧化活性的研究

中文摘要: 探究南瓜均一多糖抗氧化活性与其分子量及单糖组成之间的关系,为南瓜多糖构效关系的研究提供新参考。以葫芦科南瓜为原料,采用超声波辅助提取法提取南瓜多糖,通过Sevag 法脱蛋白及AB-8 大孔吸附树脂脱色得到除杂后的南瓜多糖(Cucurbita moschata Duch polysaccharide,CMP),经纤维素DEAE Cellulose-52 及葡聚糖凝胶Sephadex G-100 色谱柱分离纯化后得到CMP-A1、CMP-A2、CMP-B、CMP-C1、CMP-D2 5 种南瓜均一多糖,分别测定其分子量及单糖组成,并探究其与5 种均一多糖抗氧化活性之间的关系。结果表明:CMP-C1 与CMP-B 相比,两者的分子量大小相近,CMP-C1 的糖醛酸含量明显高于CMP-B,CMP-C1 对DPPH 自由基及羟基自由基的清除率比CMP-B 分别高出10.21%、11.03%;CMP-D2 与CMP-B 相比,两者的糖醛酸含量基本相等,CMP-D2 的分子量明显小于CMP-B,但CMP-D2 对DPPH 及羟基自由基的清除率仅比CMP-B 分别高出1.05%、1.96%。南瓜均一多糖的糖醛酸含量对其抗氧化活性的影响较大,糖醛酸含量越高,其抗氧化活性越强,而分子量大小对其抗氧化活性影响较小。
Abstract:This paper explored the relationship between the antioxidant activity of pumpkin polysaccharides and its molecular weight and monosaccharides composition,and provided a new reference for the study of the relationship between pumpkin polysaccharide.Pumpkin polysaccharide was extracted by ultrasonic assisted extraction with gourd pumpkin as raw material.After the decolorization of Sevag method and AB-8 macroporous adsorption resin,the decolorized pumpkin polysaccharides (Cucurbita moschata Duch polysaccharide,CMP)was obtained,5 kinds of pumpkin homogeneous polysaccharides named CMP-A1、CMP-A2、CMP-B、CMP-C1 and CMP-D2 were isolated and purified by cellulose DEAE Cellulose-52 and Sephadex G-100 chromatography column.The molecular weight and monosaccharide composition were determined respectively,and the relationship between them and the antioxidant activity of 5 kinds of homogeneous polysaccharides was investigated.The results were as follows:compared with CMP-B,the molecular weight of CMP-C1 and CMP-B was similar.The glucuronic acid content of CMP-C1 was significantly higher than that of CMP-B.The scavenging rates of DPPH·and·OH free radicals by CMP-C1 were 10.21%and 11.03%higher than that of CMP-B,respectively.Compared with CMP-B,the content of uronic acid in CMP-D2 and CMP-B was basically the same.The molecular weight of CMP-D2 was significantly less than that of CMP-B,but the scavenging rates of DPPH·and·OH free radicals by CMP-D2 were only 1.05%and 1.96%higher than that of CMP-B,respectively.The content of uronic acid in pumpkin homogeneous polysaccharide had a great influence on its antioxidant activity.The antioxidant activity increased with the increase of glucuronic acid content.and the smaller the influence of molecular weight on its antioxidant activity.
