中文摘要: 通过蛋白酶解的方法制备文蛤多肽(Meretrix meretrix L.peptide,MMP),并对其抗氧化活性进行研究。以蛋白回收率和水解度为指标,对酶解条件进行优化。得到其最佳酶解条件为,胰蛋白酶加酶量0.6%、料液比1 ∶15(g/mL)、酶解时间6 h。此时,其蛋白回收率与水解度分别为93.55%与29.55%。体外抗氧化试验结果显示,MMP 的清除DPPH自由基和羟自由基活性、以及总还原力,随浓度增加而明显增加。其中,MMP 清除DPPH 自由基活性和清除羟自由基活性的IC50 值分别为0.56、7.26 mg/mL。同时,MMP 与维生素C 在清除DPPH 自由基作用方面表现出明显的协同增效作用。在二者质量配比为10 ∶1 时,其协同增效作用达到最佳。
Abstract:Meretrix meretrix L.peptide (MMP)was prepared by hydrolysis with protease.And its antioxidant activity was analyzed.The hydrolysis conditions were optimized in order to get the highest protein recovery rate and degree of hydrolysis.On the optimum conditions,which were solid-liquid ratio 1 ∶15 (g/mL),trypsin 0.6%and enzymatic hydrolysis for 6 h,the protein recovery rate and degree of hydrolysis arrived 93.55%and 29.55%,respectively.With the increasing content,antioxidant activity of MMP obviously increased,including DPPH radical scavenging activity,hydroxyl radical scavenging activity and total reducing power.IC50 of DPPH radical and hydroxyl radical scavenging activity were 0.56 mg/mL and 7.26 mg/mL respectively.MMP and vitamin C showed significant synergistic antioxidant effect.When the ratio of MMP and vitamin C was 10 ∶1,they had the most obvious synergistic effect.
