Abstract:To compare the effects of different extraction process on the oleanolic acid content in Fructus Ligustri Lucidi and the antitumor activity of Fructus Ligustri Lucidi extract in vitro was studied.The Fructus Ligustri Lucidi was used as the raw material，and the oleanolic acid was used as the reference substance.The content of oleanolic acid was determined by ultraviolet-visible spectrophotometry，and MTT was used to detect the effect of extract on cell proliferation.The results obtained that，the four different extraction methods of oleanolic acid content in order from high to low was ethanol refluxing extraction method（13.13±0.03）mg/g，soxhlet extraction method（12.19±0.03）mg/g，ultrasonic extraction method（11.56±0.02）mg/g，cold maceration method（11.16±0.02）mg/g；The IC50 values of extracts of Fructus Ligustri Lucidi on human colon cancer cells and human liver cancer cells were 22.08，13.47 g/mL，The IC50 values of the standard oleanolic acid for human colon cancer cells and human liver cancer cells respectively were 41.45，45.90 g/mL.The results showed that the ethanol refluxing extraction had the best effect and was simple and easy to carry out；the extract of Fructus Ligustri Lucidi had inhibitory effects on human colon cancer and human liver cancer cells，and inhibited human liver cancer cells was more obvious.
