中文摘要: 研究金纳米花作为信号放大用于黄曲霉毒素M1高灵敏检测。用粒径为(91.8±0.8)nm、分支数多的金纳米花(gold nanoflowers,AuNFs)标记黄曲霉毒素M1(aflatoxin M1,AFM1)单克隆抗体(monoclonal antibody,Mab),制备用于检测牛奶中AFM1的金纳米花免疫层析检测卡(aflatoxin M1-gold nanoflower immunochromatographic test card,AFM1-GFICT)。纳米金免疫分析读数仪被用于读取AFM1系列浓度(20 pg/mL~1 000 pg/mL)的信号值。以AFM1浓度为横坐标,信号值为纵坐标绘制非线性四参数拟合曲线。非线性四参数拟合曲线相关系数R2=0.999 3,检测限为12.3 pg/mL。检测在空白牛奶样品加入AFM1标准品浓度为0.1、0.3、0.5 μg/kg 的回收率为80.4%~118.2%,变异系数(n=10)为4.27%~9.43%。15 份牛奶样品的AFM1-GFICT 和商业化AFM1-ELISA 检测结果相比,线性拟合相关系数R2=0.976 8。

Abstract:Gold nanoflowers were studied as signal amplification for high sensitivity detection of aflatoxin M1.The conditions of the monoclonal antibody(Mab)of aflatoxin M1(AFM1)labeled with gold nanoflowers(AuNFs)with a particle size of (91.8±0.8)nm and a large number of branches were optimized.Aflatoxin M1-gold nanoflower immunochromatographic test card (AFM1-GFICT)was prepared for the detection of AFM1 in milk.A nonlinear four-parameter fit curve between the AFM1series concentration (20 pg/mL~1 000 pg/mL)and its corresponding signal value was established.The correlation coefficient was R2=0.999 3 and the detection limit was 12.3 pg/mL.The recovery of blank milk samples with AFM1standard concentration of 0.1,0.3,0.5 μg/kg was measured from 80.4%to 118.2%,and the coefficient of variation(n=10)was from 4.27%to 9.43%.15 milk samples were detected by AFM1-GFICT,and the results were compared with the results of commercial AFM1-ELISA.The correlation coefficient was R2=0.976 8.
