中文摘要: 为了优化褐藻胶裂解酶酶解褐藻酸钠制备褐藻寡糖的工艺条件,在单因素试验的基础上,运用软件Design-Expert 8.0.6 设计中心组合试验,以反应体系中还原糖生成量为评价指标,采用响应面分析法确定褐藻胶裂解酶酶解的最优工艺,通过测定2,2'-联氮双-(3-乙基苯并噻唑林-6-磺酸)二铵盐[2,2'-azinobis(3-ethylbenzothi azoline-6-sulfonic acid)ammonium salt,ABTS]自由基的清除能力、抑菌指数和对对虾多酚氧化酶(polyphenol oxidase,PPO)活性的影响,检测酶解制备的褐藻寡糖的抗氧化活性和抑菌活性。结果显示最佳工艺条件:酶解时间16 h、体系pH 值8.4、酶解温度30 ℃、底物质量浓度8 g/L。此条件下,还原糖质量浓度达到1 660 mg/L。结果显示制备的褐藻寡糖具有较强的清除ABTS+自由基的能力和抑菌能力,并能有效的抑制对虾多酚氧化酶的活性。
Abstract:A simple enzymatic method for the preparation of sodium alginate oligosaccharides was investigated in this study.Response surface methodology based on single factor experiments was employed to optimized the enzymatic hydrolysis condition of alginate to obtained the maximum yield of reducing sugar and conversion of sugar.Meanwhile,the anti-oxidative activity and antimicrobial activity of alginate oligosaccharides were detected.The optimal processing condition of enzymatic hydrolysis was obtained as follows:hydrolysis time 16 h,pH value 8.4,temperature 30 ℃ ,substrate concentration 8 g/L.Under this optimal condition,the concentration of reducing sugar reached 1 660 mg/L.The alginate oligosaccharides had strong clearance power to ABTS+free radicals and antimicrobial activity,and can effectively inhibited the activity of polyphenol oxidase in shrimp.
