中文摘要: 以山楂为原料,采用响应面试验设计对离子液体酶法微波辅助提取有机酸的工艺进行优化。以离子液体种类、离子液体添加量、纤维素酶添加量、液料比和时间为自变量,山楂有机酸含量为响应值,优化得到山楂有机酸最佳提取工艺条件为:离子液体溴化-1-丁基三甲基咪唑浓度为0.039 mol/L、纤维素酶添加量质量分数1.059%、液料比29.84∶1(mL/g)、提取温度55 ℃、提取时间25.3 min,在此条件下有机酸含量为164.159 mg/g。将传统水浸泡法和离子液体酶法两种方法得到的有机酸经高效液相色谱分析,主要成分均为柠檬酸,占相对含量的88.641%和90.858%。与传统水浸泡法相比,离子液体酶法有效的缩短了提取时间。
Abstract:With hawthorn as raw material,the process of microwave assisted extraction of organic acids by ionic liquid enzyme method was optimized by response surface test.In ionic liquids,ionic liquid dosage,cellulase concentration,solid-liquid ratio and time as independent variables,organic acid content in hawthorn as response value,optimize the Hawthorn organic acid the best extraction conditions were:bromide ionic liquid Ding Jisan -1-methyl imidazole concentration was 0.039 mol/L,the adding amount of cellulase concentration was 1.059%,liquid-solid ratio of 29.84∶1 (mL/g),extraction temperature 55 ℃,extraction time 25.3 min,organic acid content under this condition was 164.159 mg/g.The organic acids obtained by the two methods of traditional water immersion method and ionic liquid enzymatic method were analyzed by high performance liquid chromatography (HPLC).The main components were citric acid,which accounted for 88.641%and 90.858%of the relative content.Compared with the traditional water immersion method,the ionic liquid enzyme method effectively shortened the extraction time.
