中文摘要: 选取5 种自行分离鉴定的乳酸菌分别对新鲜橘皮进行发酵,利用三重四极杆液质联用仪(liquid chromatograph-mass spectrometer,LC-MS)测定橘皮发酵液中9 种黄酮类成分含量(川陈皮素、桔红素、芦丁、金丝桃苷、槲皮素、木犀草素、柚皮素、芹菜素和异鼠李素),并比较发酵液中总黄酮含量,择取总黄酮含量最高的1 株作为发酵工艺优化中的发酵菌种。同时将碳源添加量、接种量、发酵时间及皮水比作为自变量,总黄酮含量作为响应值,采用中心组合试验Box-Behnken 的方法,探究各自变量及其交互作用对发酵液中总黄酮含量的影响。结果表明,通过类干酪乳杆菌(5A-1-1)发酵得到的橘皮发酵液中的总黄酮含量最高,响应面分析得到最优发酵工艺条件结果为:碳源添加量为3.25%、接种量2.97%、发酵时间2 d,皮水比1 ∶4(质量比),在此条件下获得的发酵液总黄酮含量为(0.208 667±0.008)mg/g 与预测相近,说明该响应面优化所得的最佳发酵条件可靠。

Abstract:Five kinds of Lactic acid bacteria isolated and identified by our research team were selected to ferment citrus peel.The levels of different kinds of flavonoid (naringin,tangeretin,rutin,hyperin,quercetin,luteolin,naringenin,apigenin and isorhamnetin)during fermentation process were determined by liquid chromatograph-mass spectrometer(LC-MS),also total amounts of flavonoid were calculated.The strain which produces the highest content of total flavonoids was chosen for further research. In order to optimize the total flavonoids contents from fermentation process,the effects of carbon source addition,inoculum concentration,incubation time,liquid-solid ratio and their interactions have been studied in this paper with Box-Behnken design. The results showed that the fermentation gained from Lactobacillus casei (5A-1-1)had the highest contents of flavonoid among different strains,the optimal fermentation conditions were:3.25% of carbon source addition,2.97%of inoculum concentration,2 days of incubation time and 1 ∶4 mass ratio of the solid-liquid mass ratio.Under these conditions,the total flavonoid from fermentation was(0.208 667±0.008)mg/g which was similar to that predicted.Therefore,using the response surface methodology was reliable.
