中文摘要: 研究番木瓜籽挥发油对变异链球菌UA159生物被膜形成及相关毒力基因表达的影响。采用甲基四氮盐(the abated tetrazolium salt,XTT)减低法评价番木瓜籽挥发油对变异链球菌生物被膜的抑制效果,实时定量PCR(real-time quantitative PCR,qPCR)法检测变异链球菌受番木瓜籽挥发油作用后其毒力基因luxS,gtfB,gbpD,brpA和ftf的表达情况。结果显示,番木瓜籽挥发油对变异链球菌的最低抑制生物被膜形成浓度为80 μg/mL,qPCR法检测变异链球菌的毒力基因luxS,gtfB,gbpD,brpA和ftf的表达水平受到抑制。表明番木瓜籽挥发油对变异链球菌生物被膜的形成具有抑制作用,并抑制其毒力基因luxS,gtfB,gbpD,brpA和ftf的表达。
Abstract:The antibacteria effect of Carica papaya Linn.seed essential oil on formation of UA159 biofilm and the expression of related virulence genes in Streptococcus mutans were studied.The abated tetrazolium salt was used to evaluate the inhibitory effect of Carica papaya Linn.seed essential oil on the biofilm of Streptococcus mutans.The expression levels of luxS,gtfB,gbpD,brpA and ftf genes of Streptococcus mutans after treatment with Carica papaya Linn.seed essential oil were detected by real-time quantiative PCR(qPCR).The minimum biofilm inhibitory concentrations of Carica papaya Linn.seed essential oil on Streptococcus mutans were 80 μg/mL.The results of qPCR showed that the expression levels of luxS,gtfB,gbpD,brpA and ftf genes were inhibited.The essential oil of Carica papaya Linn.seed had an obvious inhibitory effect on the formation of biofilm of Streptococcus mutans and the expression levels of luxS,gtfB,gbpD,brpA and ftf genes.
