中文摘要: 以玉米液化清液为原料,黑曲霉为发酵菌株,通过单因素筛选以及正交试验对玉米清液发酵培养基进行优化研究。通过测定产酸量及菌丝球形态的观察,最终确定最优的培养基配方为:玉米浆0.7%,(NH4)2SO41.0 g/L,MgSO40.1 g/L,CuSO40.8 mg/L,ZnSO40.02 g/L,CaCl22.0 g/L,KCl 0.1 g/L,FeSO40.34 mg/L,肌醇 0.07 g/L。通过 30 L 发酵罐放大试验确定发酵液初糖浓度为18%、种龄27 h,分段通风转速控制,发酵60 h,柠檬酸产量达到18.33 g/100 mL,糖酸转化率达到100.3%,与带渣发酵方式相比,产酸量和转化率有较大幅度提高。
Abstract:Corn liquefaction liquor was used as raw material,Aspergillus niger was used as fermentation strain,and corn liquefaction liquor fermentation medium was optimized through single-factor screening and orthogonal experiments.By observing the acid-producing and mycelial-ball morphology observations,the optimal media formulation was determined to be:0.7%corn syrup,1.0 g/L (NH4)2SO4,0.1 g/L MgSO4,0.8 mg/L CuSO4,0.02 g/L ZnSO4,2.0 g/L CaCl2,0.1 g/L KCl,0.34 mg/L FeSO4,0.07 g/L inositol.The optimum fermentation conditions were determined on 30 L scale fermenters,the optimum fermentation conditions:18%of initial sugar,the best seed culture age 27 h.The ventilation quantity and speed is controlled and fermentation time was 60 hours.The fermentation results conducted on 30 L scale fermenters showed that the citric acid production was 18.33 g/100 mL and the conversion rate of glucose reached 100.3%.It had higer level with production of citric acid from the corn liquefaction liquor fermentation than corn residue fermentation.
