中文摘要: 采用1-苯基-3-甲基-5-吡唑啉酮(1-phenyl-3-methyl-2-pyrazolin-5-one,PMP)衍生,通过高效液相色谱法(high performance liquid chromatography,HPLC)对枸杞提取物样本中单糖组成进行分析,为枸杞提取物的质量分析提供依据。自制7份枸杞多糖提取物均含有阿拉伯糖、木糖、半乳糖、葡萄糖、甘露糖、鼠李糖及半乳糖醛酸;2份市售枸杞提取物几乎只有葡萄糖,和糊精的分析结果基本一致。自制枸杞多糖的总糖含量在32.09%~43.72%之间,酸性糖含量在47.31%~57.38%之间;2份市售枸杞提取物多糖含量均大于90%,而酸性糖含量显著低于自制枸杞多糖。通过分析单糖组成、酸性糖含量能为市售枸杞提取物及相关食品和功能食品的质量控制提供依据。
Abstract:High performance liquid chromatography (HPLC)method of pre-column derivatization with 1-phenyl-3-methyl-2-pyrazolin-5-one(PMP)had been established for analysis of monosaccharide in extract of wolfberry.The analysis results showed that in seven homemade extract of wolfberry polysaccharide(WP)contained arabinose,xylose,galactose,glucose,mannose,rhamnose,and galacturonic acid.In 2 commercial WP only contained glucose which consistent with that of dextrin.The content of sugar in homemade WP was between 32.09%and 43.72%,and acidic polysaccharide was between 47.31%and 57.38%.However,in the 2 commercial WP samples(>90%)the content was significantly higher than homemade WP,acidic polysaccharide significantly lower than that of homemade WP.By monosaccharide composition analysis and determine the content of acidic polysaccharide in WP,it could provide basis for quality control of commercial WP and its related food and functional food.
