中文摘要: 建立高效液相色谱-线性离子阱质谱仪筛查和定量测定谷物中12种真菌毒素的方法。谷物样品经QuEChERS方法提取,用OasisPRiMEHLB小柱净化后,采用 ACQUITY UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8 μm),以含0.1%甲酸水溶液-含0.1%甲酸乙腈为流动相梯度洗脱分离,在电喷雾离子化正离子模式下,以多级反应监测(multi reaction monitoring,MRM)为基础,通过信息依赖性扫描方式(information dependent acquisition,IDA)触发增强子离子扫描(enhanced product ion scan,EPI)建立脱氧雪腐镰刀菌烯醇等12种真菌毒素的筛查和定量。脱氧雪腐镰刀菌烯醇等12种真菌毒素在相应的浓度范围内,线性相关性良好,相关系数(r)均大于0.995,并在相应水平下的回收率为59.5%~111.2%,相对标准偏差为0.2%~7.1%(n=6)。本方法灵敏,高效,可用于谷物中脱氧雪腐镰刀菌烯醇等12种真菌毒素的筛查与定量检测。
Abstract:Abstract:To establish a method for screening and confirming 12 kinds of mycotoxins in cereals products byhigh performance liquid chromatography-quadrupole-linear ion trap mass spectrometry.Samples were extracted by QuEChERS,purified by Oasis PRiME HLB solid phase extraction cartridge,and then were separated by gradient elution with 0.1%formic acid(A)and 0.1%formic acid acetonitrile(B)as the mobile phase using ACQUITY UPLC HSS T3 Column (2.1 mm×100 mm,1.8 μm).The analytes were determined by positive scan mode in multi reaction monitoring(MRM)mode.The linear range and linear coefficients were of mycotoxins were well.The average recoveries of the 12 mycotoxins were 59.5%-111.2%with RSD 0.2%-7.1%(n=6).This method is simple,rapid,sensitive and suitable for rapid screening and confirming of the 12 kinds of mycotoxins in cereals products.
