中文摘要: 以玫瑰花为原料,采用响应面法优化玫瑰多糖提取工艺,并对玫瑰花多糖结构及其体内抗氧化活性进行研究。结果表明:最优的提取工艺条件为提取温度为95℃,提取时间为256 min,液料比30∶1(mL/g),玫瑰花多糖得率为1.46%。紫外光谱图表明玫瑰化多糖中不含蛋白质和核酸,红外光谱显示,在3 433、2 928、1 345 cm-1等处都有很明显的多糖的特征吸收峰。抗氧化试验结果表明:玫瑰花多糖可显著提高衰老小鼠血清和肝组织中超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力(P<0.05),显著降低丙二醛(malondialdehyde,MDA)含量(P<0.01),表现出良好的体内抗氧化活性。
Abstract:The paper chooses Rosa rugosa Thunb.Petals as material.We conduct a series of studies about polysaccharide of Rosa rugosa Thunb.Petals(RRPP)about the optimization of the extraction process,preliminary structural analysis,and its antioxidant activity.The conclusions were as follows:the optimal extraction conditions for the extraction temperature:95 ℃,extraction time:256 min,liquid-solid ratio was 30 ∶1 (mL/g).Under these conditions,the RRPP extraction rate was 1.46%.Ultraviolet sepectrum analysis showed not contains nucleic acids and proteins. Infrared spectrum displayed that theRRPP have the characteristic absorption peaks(3433cm-1,2 928 cm-1and 1 345 cm-1)of polysaccharides.The antioxidant ability results showed that the activities of superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)in serum and liver tissues of the aging mice treated with RRPP significantly enhanced comparing with control mice (P<0.05),the contents of malondialdehyde(MDA) were also detected to be significantly reduced(P<0.01).So reach the conclusion that RRPP possessed a strong antioxidant activity in vivo.
