中文摘要: 以猪脑组织提取物为原料,采用硅胶柱层析法分离纯化神经节苷脂。利用单因素试验和响应面法,确定最佳纯化工艺条件:质量浓度为7.75%的样品溶液,以2.5 BV/h流速上样至装有34.89 g硅胶的层析柱后,采用乙酸乙酯与甲醇(体积比为4∶1)的混合溶剂以2.0 BV/h流速洗脱,洗脱产物中神经节苷脂的回收率为86.9%,目标化合物含量为纯化前的2.89倍,因此该纯化工艺方便可行。

Abstract:The gangliosides in pig brain tissue extract was purified by method of slica column chromatography.Based on single factor experiments and response surface methodology,the optimum conditions were as follows:the sample that gangliosides concentration of crude extract was 7.75%was loaded on the column which filled with 34.89 g slica at flow rate of 2.5 BV/h,then the eluent contained ethyl acetate and methyl alcohol,which volume ratio was 4 ∶1,washed the colume at flow rate of 2.0 BV/h,and the gangliosides in product was analysed.The recovery of gangliosides in final product was 86.9%,and the content has increased 2.89 times from the rate before purification.Therefore,the process is convenient and feasible.
