桂花黄酮类化合物体内外抗氧化活性的研究

中文摘要: 以清除DPPH·、ABTS+·的能力以及对铁离子的还原能力为指标研究桂花黄酮类化合物体外抗氧化活性;采用连续颈背部皮下注射D-半乳糖建立亚急性小鼠衰老模型作为体内抗氧化作用研究对象,评价桂花黄酮类化合物总对机体抗氧化能力的影响。结果表明,在体外试验中,以抗坏血酸作为阳性对照,一定浓度的桂花黄酮类化合物有相当的抗氧化活性;体内实验中,与空白组相比,模型组小鼠血清、肝及肾组织匀浆中丙二醛(malondialdehyde,MDA)增高,超氧化物歧化酶(superoxide dismutase,SOD)含量下降,且肝脏及肾脏中的谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、谷胱甘肽(glutathione,GSH)含量降低(P<0.05),说明亚急性衰老小鼠模型造模成功;与模型组相比,桂花黄酮高剂量组小鼠血清以及肝、肾组织匀浆中SOD、GSH、GSH-Px的活性显著增加(P<0.05),桂花黄酮各剂量组小鼠血清中MDA含量均较模型组显著降低(P<0.05),并且桂花黄酮类化合物可改善D-半乳糖致衰老小鼠肝脏组织细胞的形态结构。说明桂花黄酮类化合物在体内外具有较好的抗氧化活性,其机制与清除自由基,提高体内抗氧化酶活性和降低脂质过氧化有关。
Abstract:The antioxidant activities of total flavonoids from Osmanthus fragrans(TFOF)in vitro were evaluated by DPPH·and ABTS+·scavenging assay and the potassium ferricanide reduction methods.The aging mouse model induced by subcutaneous injection of D-galactose were the subject investigated in vivo to evaluate the effect of TFOF on the antioxidant capacity of the body.The results showed that in vitro experiments TFOF could effectively scavenge DPPH· and ABTS+·,and showed the reducing capacity on ferricyanide.And in vivo test,compared with the blank group,the malondialdehyde(MDA)contents in serum,liver and kidney homogenate of the model group were increased,and the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and glutathione (GSH)in liver and kidney were decreased(P<0.05),showed the animal aging model was successfully established by injecting D-galactose;compared with the aging group the SOD、GSH、GSH-Px content of serum、liver and kidney was increased in high dose group (P<0.05),and the content of MDA in all experiment groups was decreased(P<0.05).The TFOF could improve the cell shape and structure of D-gal aging mice's liver cells significantly.Our research showed that the total flavonoids from Osmantus fragrans have good antioxidant activity by scavenging free radicals and reducing oxidative damages.
