中文摘要: 建立高效液相色谱(High Performance Liquid Chromatography,HPLC)法对橙子中L-抗坏血酸进行含量测定研究。20 g/L偏磷酸溶液作为橙子中L-抗坏血酸的提取剂,超声5 min提取。液相色谱柱采用Inertsil ODS-3C18(5 μm,4.6 mm×250 mm),柱温 25℃,0.05 mol/L磷酸溶液-甲醇(90∶10)作为流动相,流速0.800 mL/min,检测波长 245 nm。L-抗坏血酸标准品以不同浓度进样,获得标准曲线Y=60 408.6x+38 583.7,相关系数R2=0.999 6;在1 μg/mL~50 μg/mL范围内L-抗坏血酸有良好的线性关系;检出限为0.5 μg/mL;精密度相对标准偏差RSD=0.67%。
Abstract:A method for the determination of L-ascorbic acid in orange by HPLC was established.20g/L metaphosphoric acid solution was used as the extraction agent of L-ascorbic acid in orange and ultrasonic extracted for 5 minutes.The chromatographic separation was performed on an Inertsil ODS-3C18column(5 μm,4.6 mm×250 mm),at 25℃using 0.05 mol/L phosphoric acid solution-methanol(90∶10)as the mobile phase at a flow rate of 0.800 mL/min and the detection wavelength was 245 nm.Different volume of sodium L-ascorbic acid standard were injected,the standard curve was Y=60 408.6 x+38 583.7,with a correlation coefficient of 0.999 6.It showed good linearity in the concentration range of 1 μg/mL to 50 μg/mL.The detection limit was 0.5 μg/mL.The relative standard deviation of the precision was 0.67%.
