猕猴桃中甲基硫菌灵和异菌脲HPLC测定方法研究

中文摘要: 建立高效液相色谱法(High Performance Liquid Chromatography,HPLC)同时测定猕猴桃中甲基硫菌灵和异菌脲两种农药的含量。猕猴桃样品用乙腈涡旋提取,采用反相高效液相色谱法测定,色谱柱为Diamonsil C18柱(250 mm×4.6 mm,5 μm),柱温为 30℃,进样体积 20 μL,流速 1.0 mL/min,检测波长 210 nm,流动相为乙腈-0.05%甲酸水溶液=(60 ∶40),采用等度洗脱模式。结果表明,甲基硫菌灵和异菌脲分别在 0.204 μg/mL~10.2 μg/mL 和 0.210 μg/mL~10.5 μg/mL范围内线性关系良好,方法的专属性和准确度良好,能够用于检测猕猴桃样品中上述两种农药的残留。
Abstract:A High Performance Liquid Chromatography(HPLC)method was established for the simultaneous determination of thiophanate-methyl and iprodione in kiwifruit.The optimal method was applied with Diamonsil C18column (250 mm×4.6 mm,5 μm),acetonitrile ∶0.05%methanoic acid solution=60 ∶40 as the mobile phase,the flow rate was 1.0 mL/min,the column temperature was maintained at 30 ℃ and the UV detection wavelength was set at 210 nm.The linear relationship between thiophanate-methyl and iprodione concentration and peak area were good in the range of 0.204 μg/mL-10.2 μg/mL (r=0.999 6)and 0.210 μg/mL-10.5 μg/mL(r=0.999 8),respectively.The method is reliable,sensitive,rapid and can be used for determination of thiophanate-methyl and iprodione in Kiwifruit.
