大孔树脂纯化酸浆果实多酚及其生物活性研究

中文摘要: 以酸浆果实为研究对象,多酚经乙醇浸提,采用9种大孔吸附树脂对酸浆果实多酚进行初步纯化,利用化学方法对酸浆果实多酚的抗氧化活性以及对酶活性的抑制作用进行了研究。结果表明AB-8树脂纯化酸浆果实多酚效果较好,最佳纯化条件为:样液质量浓度0.3 mg/mL、pH为样品本身的pH值、上样流速1 mL/min、乙醇体积分数为80%,洗脱流速为1 mL/min。酸浆果实多酚具有一定还原能力,对OH自由基和DPPH自由基的IC50分别为0.604 4 mg/mL和2.262 mg/mL;并对胰脂肪酶、α-淀粉酶和α-葡萄糖苷酶有抑制作用,其IC50值分别为0.826 2 mg/mL、0.817 8 mg/mL和1.011 mg/mL。本研究表明酸浆果实多酚具有一定的体外抗氧化活性,并对胰脂肪酶、α-淀粉酶和α-葡萄糖苷酶的活性有抑制作用。
Abstract:Using the fruits of Physalis alkekengi as the research object,polyphenols was extracted using alcohol.Nine macroporous resins were selected to purify polyphenols of the fruits of Physalis alkekengi and the optimal purification conditions,and its antioxidant activity and inhibition of enzyme activity were determined by using chemical methods.The results showed that the AB-8 macroporous resin had the best purification effect;and the optimal purification conditions were as follows:polyphenols concentration in a loading solution was 0.3 mg/mL;the optimal pH value was the pH value of the sample itself;the flow rate of the column was 1 mL/min;the ethanol concentration was 80%;and the elution flow rate was 1 mL/min.The experimental results showed that the Physalis fruit polyphenols had a certain reducing capacity on·OH and DPPH·,with the scavenging IC50values were 0.604 4 mg/mL and 2.262 mg/mL.The IC50for inhibiting activity against pancreatic lipase,α-amylase and α-glucosidase were 0.8262 mg/mL,0.817 8 mg/mL and 1.011 mg/mL,respectively.The results demonstrated that the Physalis fruit polyphenols have antioxidant activity and inhibitory effects against pancreatic lipase,α-amylase and α-glucosidase in vitro.
