中文摘要: 以拐枣为试验材料,采用酶法对可溶性膳食纤维进行提取。在酶解温度、酶解时间、pH值、酶用量4个单因素试验基础上,利用Box-Behnken的中心组合设计原理进行响应面设计优化提取工艺参数。结果表明:拐枣中可溶性膳食纤维(soluble dietary fiber,SDF)得率最佳提取工艺条件为酶解温度60℃,酶解时间70 min,pH值为5.0,酶用量0.6%。拐枣中SDF得率可达14.22%,与SDF的提取理论值比较,其相对误差约为0.42%,且重复性好,说明响应面法建立的数学模型对拐枣中SDF提取具有稳定可靠性。
Abstract:The soluble dietary fiber were extracted from Hovenia dulcis with the enzymolysis and response surface methodology.Based on the single factor experiments,Box-Behnkencentral composite design principles were used to select enzyme temperature,pH,enzyme time and dosage as four parameters of response surface design to optimize extraction process.The results showed that the optimum extraction conditions of the extraction rate of Hovenia dulcis SDF enzyme temperature 60 ℃ ,pH 5.0,enzyme time 70 min and enzyme dosage 0.6%.The extraction rate of soluble dietary fiber reached 14.22%,compared with the theoretical value,the relative error was about 0.42%,and the repeatability was also very good,that was to say using the model of response surface extracting the Hovenia dulcis SDF was stability and reliability.
