中文摘要: 在柠檬酸作用下,以人参须根粉为原料直接提取制备人参皂苷Rg5,以达到简化试验步骤、节省时间、绿色制备的目的。通过单因素试验探索提取温度、酸浓度和固液比对人参皂苷Rg5得率的影响,响应曲面分析法对人参皂苷Rg5的提取制备工艺进行优化,通过高效液相色谱法(HPLC)对人参皂苷Rg5进行定量分析。试验结果表明,人参皂苷Rg5的加样回收率为99.73%~101.72%,平均加样回收率为100.37%;人参皂苷Rg5以在溶液中48 h内稳定性良好;在柠檬酸的催化下,水-正丁醇=8∶2(体积比)的混合溶剂为提取剂,提取温度为112℃,酸浓度为0.14 mol/L,固液比为1∶28(g/mL),人参皂苷Rg5得率为1.54%。

Abstract:To direct extraction of ginsenoside Rg5 from ginseng fibrous root powder,using citric acid as catalyst,ginseng fibrous root powder(GFRP)as raw material.The preparation process was optimized by response surface method basised on single factor experiment,the extraction temperature,acid concentration and solid to liquid ratio were investigated,and the rate of ginsenoside Rg5 was analyzed by HPLC.The results showed that the recovery rate was 99.73%-101.72%and the average recovery rate was 100.37%;ginsenoside Rg5 had good stability within 48 h in solution;and when the volume rate of water:n-butanol was 8∶2,the extraction temperature was 112℃,the acid concentration was 0.14 mol/L,the solid-liquid ratio was 1∶28(g/mL),the extraction rate of ginsenoside Rg5 could reach 1.54%.
