中文摘要: 探讨蒽酮-硫酸法测定多糖含量的影响因素,并优化榛花多糖含量测定的最佳条件。试验以吸光度值为评价指标,检测波长为625 nm,对蒽酮质量浓度、加热温度、蒽酮试剂用量和加热时间进行单因素试验,再用L9(34)正交试验设计优选测定条件。结果表明,1 mL样液下,蒽酮质量浓度20 mg/mL、加热温度90℃、蒽酮试剂用量4.0 mL、加热时间10 min的条件最优。最佳工艺下,平均回收率为99.63%,相对标准偏差(RSD)为1.58%。

Abstract:To optimize the content determination conditions ofCorylus hecerophyllapolysaccharide by anthrone-sulfuric acid method.Anthrone-sulfuric acid method was used for content determination glucose as parameters.The absorbance was measured at 625 nm.A single factor test was performed on the concentration,heating temperature,anthrone reagent dosage and heating time.L9(34)orthogonal design was chosen for optimum measuring conditions.The results showed that 10 min reaction at 90℃in the presence of 4.0 mL of 20 mg/mL anthrone reagent was found to be optimal in 1mL sample fluid.The average recovery was up to 99.63%,and the RSD was 1.58%.
