中文摘要: 以香菇液体发酵胞外粗多糖得率为主要评价指标,通过正交试验研究液体发酵最优培养基配方;测定胞外 和胞内粗多糖中蛋白质、还原糖和多糖含量,并研究其体外对羟自由基和 DPPH 自由基的清除率。结果表明:香菇液 体发酵最优配方为葡萄糖 9 g、酵母膏 0.9 g、可溶性淀粉 6 g、麦麸皮 24 g。此配方下香菇胞外粗多糖中蛋白质含量0.68 %,还原糖含量 6.17 %,多糖含量 25.6 %;胞内粗多糖中蛋白质含量 1.16 %,还原糖含量 7.01 %,多糖含量 32.3 %; 胞外粗多糖对羟自由基清除率为 63.05 %,对 DPPH 自由基清除率为 62.05 %。
Abstract:The extracellular polysaccharide yields of Lentinus edodes liquid fermentation was took as main eval- uation index to determine the optimal formula of liquid fermentation medium. The protein, reducing sugar and polysaccharide content of extracellular and intracellular polysaccharide was also studied. Then its clearance rate to hydroxyl radical (·OH)and DPPH·was studied in-vitro. The results showed that the optimal formula of L. edodes liquid fermentation contains glucose 9 g, yeast extract 0.9 g, soluble starch 6 g, wheat bran 24 g. We found that the protein content in extracellular polysaccharide of L. edodes accounts for 0.68 %, reducing sugar content was 6.17 %, polysaccharides content was 25.6 %; protein content in intracellular polysaccharide was 1.16 %, reducing sugar content was 7.01 %, polysaccharides content was 32.3 %. The clearance rate of extra- cellular polysaccharide to hydroxyl radical was 63.05 %, to DPPH free radicals was 62.05 %.
