中文摘要: 研究木瓜蛋白酶酶解法提取林蛙卵黑色素工艺,分析林蛙卵黑色素的抗氧化活性。以林蛙卵为原料,通过单 因素试验和正交试验,分析木瓜蛋白酶的用量、料液比、酶解温度、酶解时间对黑色素提取率的影响,确定林蛙卵黑色 素酶水解法的最佳提取工艺;应用 DPPH 法测定林蛙卵黑色素对 DPPH·的清除能力。得出木瓜蛋白酶解法提取林蛙 卵黑色素的最佳条件为:酶用量为 30 mg,反应温度 58 ℃,料液比 1 ∶ 20(g/mL),反应时间 4 h,林蛙卵中黑色素的提取 率为 13.23 %。黑色素对 DPPH·具有显著的清除作用,0.4 mg/mL 的林蛙卵黑色素对 DPPH·的清除率达 52 %。

Abstract:This study focused on the process of melanin extraction from Rana temporaria chensinensis eggs by papain hydrolysis, and analyzed the antioxidant activity of extracted melanin. Using Rana temporaria chensi- nensis eggs as material, the effects of papain concentration, material to liquid ratio, hydrolysis temperature and hydrolysis duration were studied by single factor experiments and orthogonal test analysis, determined the opti- mum hydrolysis process. The scavenging activity on DPPH·of the extracted melanin was measured by DPPH method. The optimum hydrolysis parameters for extraction of melanin from Rana temporaria chensinensis frog eggs was as follows: enzyme dosage 30 mg, hydrolysis temperature 58 ℃, material to liquid ratio 1 ∶ 20(g/mL), hydrolyze duration 4 h. The yield of melanin was up to 13.23 % by this process. The melanin has obvious clear- ance ability on DPPH·. The ability of melanin for DPPH·scavenging at 0.4 mg/mL was 52 %.
