中文摘要: 以 RID 为检测器,建立高效液相色谱法定量测定发酵液中葡萄糖和 D-核糖的方法。该方法的测定条件为: 柱型 Shodex Suger SH1011,流动相为 0.005 mol/L 的稀硫酸溶液,柱温为 50 ℃,进样量为 10 μL。底物葡萄糖和产物 D核糖的出峰时间分别为 11.573 min 和 13.313 min。 得到底物葡萄糖和产物 D-核糖的回归方程分别为 Y=6.868 4X-0.069 1 和 Y=6.556 5X+0.004 6,其中 Y 为葡萄糖或 D-核糖的质量浓度,X 为对应的峰面积。线性范围在 2 g/L~45 g/L 时,该法精密度、稳定性和重现性均良好;葡萄糖的平均回收率为 99.27 %,D-核糖的平均回收率为 99.61 %,RSD 值 分别为 0.38 %和 0.14 %,说明本方法准确可行。
Abstract:A rapid HPLC method for simultaneously and quantitatively determining the D-ribose and glucose with RID-detector was established as: Shodex Suger SH1011 column, 0.005 mol/L of diluted sulfuric acid solu- tion as the mobile phase, column temperature of 50 ℃ and the injection volume was 10 μL. Retention time of glucose and D-ribose was 11.573 min and 13.313 min, respectively. The regression equations of glucose and D-ribose were Y=6.868 4X-0.069 1 and Y=6.556 5X+0.004 6, respectively, in which Y means concentration of glucose/D-ribose and X means the peak area ratio. Within the linear range of 2 g/L-45 g/L, the precision, sta- bility and reproducibility of the established method was good. The average coefficient of recovery of glucose and D-ribose were 99.27 % and 99.61 %, RSD were 0.38 % and 0.14 % respectively, which revealed that the method was accurate and credible.
