中文摘要: 以苦荞麦为原料,通过碱提酸沉法提取蛋白质复合物,并利用单因素试验和响应面软件优化最佳蛋白质提 取工艺,优化结果为提取时间 50 min,料液比 1 ∶ 12(g/mL),提取温度 55 ℃,pH 10.0。正己烷去除苦荞麦蛋白质复合物 中的黄酮类化合物和脂类。碱性蛋白酶酶解苦荞麦蛋白制备酶解液,研究不同水解度的酶解液的总抗氧化能力,超氧 阴离子自由基、羟自由基、DPPH 自由基的清除能力,结果表明不同水解度的苦荞蛋白酶解液都具有抗氧化能力。但 具有不同的自由基清除活力,其中超氧阴离子自由基的清除活性最大,其次是 DPPH 自由基清除活性,几乎不具有羟 自由基清除活性。
Abstract:Fagopyrum tataricum was used as raw material, then using alkali extraction and acid precipitation method to extract protein complex, using the single factor experiment and response surface software to optimize the best protein extraction technology, the optimization results was extracting time 50 min, olid-liquid ratio 1 ∶ 12(g/mL), extraction temperature 55 ℃, pH 10.0. Removed the flavonoids and lipids in tartary buckwheat protein by n-hexane. Then, hydrolyzed tartary buckwheat protein into polypeptide solution by alkaline pro- tease. So we can research on the total antioxidant capacity, superoxide anion radical, hydroxyl free radical and DPPH radical scavenging activity of enzymatic hydrolysate with different degree of hydrolysis. The result showed that the enzymatic hydrolysates above,all had antioxidant ability. But they had the different radical scavenging activity, the best was followed by DPPH·and nearly had no·OH scavenging activity.
