中文摘要: 建立一种快速、有效的高效液相色谱法测定运动食品中纳他霉素残留的方法。样品经甲醇提取后,采用 HLB 固相萃取小柱进行净化和富集。采用 Waters RP C18 色谱柱(4.6 mm×250 mm,5 μm)分离,以甲醇 ∶ 水 ∶ 乙酸(体积比)=60 ∶ 35 ∶ 5 作为流动相,用 PDA 检测器检测,外标法峰面积定量。结果表明,纳他霉素标准在 0.50 μg/mL~10.00 μg/mL 浓度范 围内线性良好,相关系数 r2 为 0.9999,检出限为 10.0μg/kg,定量限为 30.0μg/kg,加标回收率达到 84.6 %~90.5 %。该方法具有 前处理简单、净化效果好、灵敏度高和检测速度快的优点,适用于运动食品中纳他霉素残留的分离和定量检测。
Abstract:A rapid and effective method was established for the determination of natamycin in sports food by HPLC. After the sample was extracted by methanol, it was concentrated and purified by HLB solid phase ex- traction column. The separation of target compound was performed on a Waters RP C18 chromatographic column (4.6 mm×250 mm, 5 μm) using methanol ∶ water ∶ acetic acid(60 ∶ 35 ∶ 5)as mobile phase, with PDA detector and external standard method peak area quantification. The linear range of natamycin was in the range of 0.50 μg/mL-10.00 μg/mL with a correlation coefficient of 0.999 9. The detection limit was 10 μg/kg and the quantitative limit was 30.0 μg/kg.The recovery rate was 84.6 %-90.5 %. The method has the advantages of sim- ple pretreatment, good purifying effect, high sensitivity and fast detection rate. It is suitable for the separation and quantitative detection of natamycin in sports food.
