中文摘要: 以磷酸盐缓冲液匀浆浸提法提取混合甘蔗汁多酚氧化酶(PPO),探讨缓冲液 pH 值、表面活性剂添加量和聚乙 烯吡咯烷酮(PVPP)添加量对甘蔗汁 PPO 提取效果的影响。结果表明,以邻苯二酚为底物时,获得甘蔗汁 PPO 的酶活性 最适测定波长是 230 nm。磷酸盐缓冲液提取甘蔗汁 PPO 最适 pH 值为 6.2;在磷酸盐缓冲溶液中 PVPP 最适添加量为2 %,提取甘蔗汁 PPO 的比活性最高。在缓冲液中加入表面活性剂 Tween 80 和 Triton X-100 时,提取甘蔗汁 PPO 的比活 性较低。

Abstract:Polyphenol oxidase (PPO) was extracted from mix sugarcane juice by phosphate buffer homogenate extraction. The effects of pH value, surfactant adding quantity and polyvinyl polypyrrolidone (PVPP) adding quantity on the PPO extraction were determined. The results showed that the optimal wavelength of enzymatic activity of PPO was 230 nm when catechol was used as substrate. The optimal pH value of PPO was 6.2. The op- timal adding quantity of PVPP was 2 % in phosphate buffer solution, and the highest specific activity of PPO was obtained in sugarcane juice. When adding surfactant Tween 80 and Triton X-100 in buffer, the specific ac- tivity of PPO in extracting sugarcane juice was low.
