3种植物活性成分联合改善2型糖尿病小鼠 氧化应激效果研究

中文摘要: 建立 2 型糖尿病小鼠模型,测定 FBG、TG、TC、HDL、LDL、INS、FFA、MDA、ROS 等,分析南瓜多糖、葛根黄酮、普 洱茶褐素提取物单一及联合对糖脂代谢和氧化应激的作用效果及氧化应激与糖脂代谢的相关性。研究表明提取物单一 及联合干预均对糖脂代谢和氧化应激指标有显著的改善(P<0.05)。其中多糖与黄酮联合组对改善 FFA、MDA 的效果显 著优于多糖组、黄酮组,对改善 TG 的效果显著优于多糖组(P<0.05);多糖与茶褐素联合组较多糖组可显著降低 TG、 LDL、INS 的水平(P<0.05);茶褐素与黄酮联合组对 TG、TC、LDL、FBG、INS、FFA 的作用效果显著优于黄酮组,对 TG 的作 用效果显著优于茶褐素组(P<0.05)。利用主成分分析可知,联合与单一成分在改善糖、脂代谢及氧化应激指标上权重贡 献率不尽相同,说明其作用机制的靶点和通路存在差异性。同时,通过线性相关分析法可知氧化应激与糖脂代谢具有极 显著的正相关关系,提取物对 T2DM 小鼠糖脂代谢的改善与降低氧化应激水平有关。
Abstract:In this study, the type 2 diabetes model was established and FBG,TG,TC,HDL,LDL,INS, FFA, MDA,ROS were detected to analyze the effect of pumpkin polysaccharide, pueraria flavonoids and pu -erh theabrowns on oxidative stress, glycolipid metabolism and its relationship. The study showed that the single and combined intervention of each extract had significant effects on glycolipid metabolism and oxidative stress (P <0.05). Subsequently, the indicators for oxidative stress and glycolipid metabolism were screened through the PCA analysis, the results showed that of the ratio of weight dedication of individual extracts and combinations were different, and the data suggested that pumpkin polysaccharide, pueraria flavonoids , pu-erh theabrowns single and combined extracts have different oxidative stress and glycolipid metabolism target points and path- way. Furthermore, the linear correlation analysis showed that the oxidative stress and glycolipid metabolism have a significant positive correlation, and the improvement on glycolipid metabolism is related with the level of oxidative stress in type-2 diabetic mice .
