中文摘要: 主要研究培养条件对动物双歧杆菌活菌数的影响,design-expert 软件辅助优化设计, 以期更好的为双歧杆菌的研 究提供基础数据及满足工业化生产需要。采用改良的玉米糖化液为培养基,以活菌数为响应值,选取不同的玉米糖化液浓 度、培养温度、接种量和初始 pH,借助 design-expert 软件建立四因素三水平的试验,得到响应值与各因素之间交互作用的 数学模型。结果表明:接种量与培养温度一次项显著,4 个因素的二次项及玉米糖化液浓度与接种量的交互项显著,其他项 不显著。动物双歧杆菌的最佳培养条件分别为:接种量 7.4%,玉米糖化液浓度 12.0°Bx,初始 pH6.5,培养温度 37.4℃。

Abstract:The research mainly explored optimization of fermentation conditions for Bifidobacterium animalis by response surface methodology,based on design-expert software design,in order to provide the basis data for the further research and meet the factory need. Meanwhile,the contact of Bifidobacterium animalis, corn juice con- centration,pH and fermentation temperature were investigated on viable count. The mathematical model of four factors and three levels was created according to design-expert software design and Box -Behnken experimental design. The result showed that the interaction term of the contact of Bifidobacterium animalis and fermentation temperature and individual quadratic interaction term are significant,but others are unsignificant. The optimum culture conditions were obtained: 7.4 % of the Bifidobceterium animalis,12.0 °Bx of the corn juice concentra- tion,pH 6.5,fermentation temperature of 37.4 ℃.
