中文摘要: 通过试验确定 RP-HPLC 法分析芽孢皮层肽聚糖水解物的最佳条件如下: 在 Welch Ultimate AQ-C18 色谱柱 上,柱温为 30 ℃,流速为 1 mL/min,检测波长为 206 nm,洗脱时间为 120 min,以甲醇和 0.05 mol/L 磷酸盐缓冲液(pH=4.31)做流动相,在 0 min~5 min 时使用 100 %磷酸盐缓冲溶液,在 5 min~120 min 时流动相中的甲醇浓度从 0 梯度增 加到 30 %。结果表明:用 RP-HPLC 法分析芽孢皮层肽聚糖水解物是可行的,为研究高压热处理下芽孢皮层肽聚糖支架 水解机理,并进一步阐明高压热处理杀灭细菌芽孢的机理提供了研究方法支撑。

Abstract:By experiment, the optimal RP-HPLC conditions for analyzing spore's cortical peptidoglycan hy- drolysate were as follows: on Welch Ultimate AQ-C18 chromatographic column, column temperature 30 ℃, flow speed 1 mL/min, detection wavelength 206 nm, elution time 120 min, with methanol and 0.05 mol/L PBS (pH = 4.31)as mobile phase, in 0 min-5 min 100 % PBS was used, in 5 min-120 min the methanol concentra- tion increased gradually from 0 to 30 %. The result indicated that it was feasible to use RP-HPLC for analyzing spore's cortical peptidoglycan hydrolysate, which provided research method support for studying the hydrolysis principle of spore's cortical peptidoglycan under HPTS (High Pressure Thermal Sterilization) treatment, and further for clarifying the principle of spore inactivation by HPTS.
