中文摘要: 采用微波-超声波协同提取落葵中的多糖,研究超微粉碎与普通粉碎的多糖提取率差别,并对超微粉碎的提取 进行条件优化。结果表明:1)在设定的相同条件下,超微粉碎落葵的多糖提取率比普通粉碎提高 64.11 %;2)通过单因素 试验考察了液料比、时间、超声波功率和微波功率对于超微粉碎落葵多糖提取率的影响;3)在单因素试验的基础上,采 用 Box-Behnken 中心设计对超微粉碎落葵进行微波-超声波协同提取多糖条件优化, 确定最佳工艺条件为微波功率 331 W、超声波功率 193 W、提取时间 18 min、液料比 50 mL/g,在此最佳工艺条件下,落葵多糖提取率的可达到 27.39 %, 相对误差为-2.35 %,与理论值基本符合,表示该模型能很好的预测实际提取情况。

Abstract:Polysaccharide was extracted from Basella rubra L. using microwave -ultrasound assisted method. The difference of influence of ultramicro grinding and ordinary grinding on extraction rate was studied. And the ultramicro extraction conditions were optimized as well. The results demonstrated that: (1)there was 64.11 % increased in polysaccharide extraction rate for ultramicro grinding than that of the normal grinding technology under the same conditions; (2)The influence of liquid material ratio, time, ultrasonic power and microwave power on the polysaccharide extraction ratio from Basella rubra L. were investigated through the single factor experiment; (3)Box-Behnken Center design was used on the basis of single factor test to optimize the mi- crowave-ultrasonic assisted extracting conditions of polysaccharide. The optimum conditions were determined as the microwave power was 331 W, ultrasonic power was 193 W, extraction time was 18 min, and the liquid-sol- id ratio was 50 mL/g. The extraction rate could reach 27.39 % and the relative error was -2.35 % under the opti- mum conditions, which was consistent with the theoretical value. So the model was proved to be effective to pre- dict the actual extraction rate.
