不同品系猴头菇子实体中总酚含量测定

中文摘要: 该文以猴头菇子实体为原料,采用超声波辅助提取法优化猴头菇总酚提取工艺条件。采用 Folin-Ciocalteu 比 色法测定总酚的含量,没食子酸为标准品,以猴头菇子实体中总酚的含量为评价指标,分别对提取溶剂、提取时间和 料液比进行优化,建立了猴头菇子实体中总酚的含量测定方法,得到最佳料液比 1 ∶ 40(g/mL),纯水提取,提取时间 60 min,总酚含量在 0~14 μg 时与吸光度呈良好的线性关系,其回归方程 Y = 0.037 3X+0.003 7,r ≥ 0.999,平均加样回 收率为 101.41 %,RSD 为 1.20 %。同时,将该方法在最适条件下测定了 10 个不同品系猴头菇子实体中总酚的含量,不 同品系猴头菇子实体中总酚的含量在 2.20 mg/g~4.77 mg/g 之间。结果表明,该方法简单、快速、重复性好,可用于猴头 菇子实体的总酚含量测定方法。
Abstract:The total polyphenol content from Hericium erinaceus was abstracted by ultrasonic-assisted extrac- tion . The total polyphenol content was analyzed by Folin-Ciocalteu(FC) colorimetry method, and gallic acid was used as reference substance. Effects of extraction solvent, extraction time and solid to liquid ratio on total polyphenols content from Hericium erinaceus were investigated. It showed that the optimized extraction condi- tions were water extract, the extraction time was 60 min and the solid-liquid ratio is 1 ∶ 40(g/mL). The calibra- tion curves were linear over gallic acid content ranging from 0~14 μg, the typical standard curves were Y =0.037 3X+0.003 7 with the line regression correlation coefficients greater than 0.999. The average recovery was 101.41 % with RSD of 1.20 %. This method was also successfully applied to the determination of total polyphe- nols content from ten different Hericium erinaceus strains. The total polyphenol content from different Hericium erinaceus strains between 2.20 mg/g~4.77 mg/g .This method established is simple, rapid and repeatable, which is suitable for analysis of total polyphenols in Hericium erinaceus.
