中文摘要: 提取纯化枇杷叶多糖,并对其组分进行研究。枇杷叶多糖提取液经大孔树脂除杂,醇沉后干燥即得枇杷叶可 溶性总多糖。经 DEAE-52 纤维素柱层析及 Sephadex G-200 葡聚糖凝胶柱层析,得到纯化后多糖,并采用 Sephadex G-200 葡聚糖凝胶柱测定多糖的分子量, 水解多糖后采用薄层层析对枇杷叶多糖进行组分鉴定及单糖的组成比例测定。 通过 DEAE-52 柱层析得 3 个枇杷叶多糖酸性组分, 分别经过 Sephadex G-200 柱层析得到 3 个乳白色组分, 分别为 PSL-1、PSL-2、PSL-3。3 个组分的洗脱曲线均为单一对称峰, 初步判断 3 个组分含有单一成分的多糖。PSL-1、PSL-2、 PSL-3 分子量分别约为 700、500、400 kDa。 经过薄层层析初步推断出 PSL-1 含有鼠李糖和葡萄糖, 含量之比为 1 ∶ 0.602; PSL-2 含有葡萄糖、半乳糖、鼠李糖,含量之比为 1 ∶ 1.415 ∶ 1.408;PSL-3 含有半乳糖和鼠李糖,含量之比为 1 ∶ 1.03。
Abstract:Polysaccharides of loquat leaf was extracted and purified, and its composition was studied. The total soluble polysaccharide of loquat leaf was prepared from the crude extract which was primary purified by macrop- orous resin and then precipitated by adding alcohol. Pure polysaccharides were obtained by DEAE-52 cellulose column chromatography and Sephadex G-200 gel column chromatography, and then the molecular weights of polysaccharides were determined by Sephadex G-200 gel column chromatography. The polysaccharides were hydrolyzed, and then the monosaccharide component and ratio were identified by thin layer chromatograph. Three fractions were obtained by DEAE-52 column chromatography, and then three components were obtained through Sephadex G-200 column chromatography that confirmed to be single component based on single sym- metrical pea of the elution curves respectively, named PSL-1, PSL-2 and PSL-3. The molecular weight of PSL-1, PSL-2 and PSL-3 were 700,500,400 kDa respectively. The monosaccharide component and ratio of the three polysaccharides of loquat leaf were identified by thin layer chromatograph. PSL-1 contains rhamnose and glucose, and the ratio was 1 ∶ 0.602. PSL-2 contains glucose, galactose and rhamnose, and their ratio was 1 ∶ 1.415 ∶ 1.408. PSL-3 contains galactose and rhamnose, and their ratio was 1 ∶ 1.03.
