中文摘要: 以百令胶囊及野生冬虫夏草作为对照品, 优化了 HPLC 法测定冬虫夏草菌 F4539 指纹图谱的色谱条件,并 计算其中 4 种核苷成分的含量。结果表明:采用 YMC-Pack Pro C18(4.6 mm×250 mm, 5 μm)色谱柱,甲醇 ∶ 水(20 ∶ 80, 体积比)为流动相,流速 0.6 mL/min,检测波长为 260 nm 时,待测样品得到较好的基线分离;冬虫夏草菌 F4539 发酵 菌丝体及发酵液中含有与野生冬虫夏草及百令胶囊相同的有效成分;4 种核苷物质在 2、4、6、8、10 mg/L 的浓度梯度 下,线性关系良好;通过定量计算,冬虫夏草菌 F4539 菌丝体及发酵液中的腺苷含量分别为 0.094 mg/g 和 2.53 mg/g, 百令胶囊中腺苷含量为 1.45 mg/g。本研究对于冬虫夏草 F4539 的功效评价、发酵后处理及产业化开发提供了重要技 术参数。

Abstract:Taking Bailing capsule and wild Cordyceps sinensis as reference substance, the HPLC chromatogra- phy conditions for detecting of Cordyceps sinensis F4539 fingerprint chromatography was optimized, and the content of four kinds of nucleotide substance was calculated. The HPLC conditions were YMC-Pack Pro C18 column(4.6 mm×250 mm, 5 μm), methanol ∶ water(20 ∶ 80, volume ratio)as a mobile phase, flow rate 0.6 mL/min, determine wavelength, 260 nm; the samples could be separated well; the Cordyceps sinensis F4539 fermenta- tion mycelia and broth contained the same active ingredients with wild Cordyceps sinensis and Bailing capsule; under the concentration gradient of 2,4,6,8,10 mg/L, a good linear relationship was showed between peak area and concentration for four kinds of nucleoside substance;through quantitative calculation, the content of adenosine from the Cordyceps sinensis F4539 mycelia and fermentation broth were respectively 0.094 mg/g and 2.53 mg/g, the content of adenosine was 1.45 mg/g for Bailing capsule. This research provided an important technical parameters for the efficacy evaluation of Cordyceps sinensis F4539, fermentation processing and in- dustrialization development.
