中文摘要: 以石榴皮为研究对象,用毒害小的乙醇溶液为提取溶剂,以安石榴苷为分离目标,用高效液相色谱法检测安 石榴苷含量,采用 Box-Behnken 中心组合设计原理设计试验,应用响应面法优化提取工艺条件;通过考察·OH、O2-·、DPPH·清除能力,评价提取物的体外抗氧化性。响应面法优化结果显示,安石榴苷提取的最优条件为:乙醇浓度 60 %, 料液比 1 ∶ 25(g/mL),提取温度 60 ℃,提取时间 6 h,在此条件下进行验证,安石榴苷的提取量可达到 70.89 g/kg,各因 素对安石榴苷提取的影响顺序为:乙醇浓度>提取温度>提取时间>料液比;自由基清除试验显示,安石榴苷粗提物可 以较好地清除·OH、O2-·、DPPH·,且对各自由基清除能力明显高于石榴皮原料。
Abstract:The pomegranate husk was the research subject, and the ethanol was used as extraction solvent, which is less toxic. Punicalagin was extracted as object from the pomegranate husk, and was tested by HPLC. Response surface methodology was used to optimize the extraction condition, and Box-behnken principle was used to design the test. By studying the scavenging rate of·OH,O2-·,DPPH·, the antioxidation in vitro of the punicalagin extract was assayed. The result of the optimization test of response surface methodology showed that the optimized condition for punicalagin extraction was: the content of ethanol solution was 60 %, the ratio of po- mace to ethanol solution was 1 ∶ 25(g/mL), and temperature was 60 ℃and time were 6 h, and in this condition the extracted punicalagin quantity could reach to 70.89 g/kg, and the sequence of each factor effecting on ex- tracted punicalagin quantity was: the content of ethanol solution> temperature> time> the ratio of material to so- lution; the free radical scavenging test showed that the punicalagin extract could scavenge·OH,O2-·,DPPH·very well, and exhibited better capacity of free radicals scavenging than the pomegranate husk.
