阿魏菇多糖超声微波辅助提取及其抗氧化活性

中文摘要: 以阿魏菇作为原料,水作为提取溶剂,通过响应面优化超声-微波协同辅助提取阿魏菇多糖工艺,并和传统水浴浸提法进行比较,采用清除DPPH·、·OH和O2-·模型对其体外抗氧化活性进行评价。结果表明:超声-微波辅助提取阿魏菇多糖的最佳的工艺条件为:料液比1∶50(g/mL),提取时间10min,微波功率60W。与传统水浴浸提法相比,超声-微波辅助提取缩短了提取时间,阿魏菇多糖的得率由2.23%增加到5.6%。超声-微波协同辅助提取对阿魏菇多糖的结构基本没有影响。阿魏菇多糖具有较强的清除DPPH·、·OH和O2-·的能力,并与质量浓度呈一定正相关关系,当阿魏菇多糖质量浓度达到5mg/mL时,对DPPH·、·OH和O2-·的清除率分别达到67%、59%和63%,但弱于VC的抗氧化活性。
Abstract:Pleurotus ferulae was used as raw material.Water was used as extraction solvent.Ultrasonic microwave assisted extraction(UMAE)technology of polysaccharides from Pleurotus ferulae(PFP)was optimized by response surface methodology.The traditional water bath extraction(TWBE)was compared with UMAE.Antioxidant activity of PFP in vitro was evaluated by hydroxyl,superoxide anion and DPPH free radical scavenging method The optimum technology condition for ultrasonic microwave assisted extraction of PFP was determined as follows:the material-liquid ratio was 1∶50(g/mL),the extraction time was 10 min and microwave power was 60 W.Compared with the traditional water bath extraction,the ultrasonic microwave assisted extraction was time saving and increased the yield from 2.23%to 5.6%.UMAE had no effect basically on structure of PFP.PFP had strong scavenging effects of DPPH radical,hydroxyl radical and superoxide anion radical.Scavenging effects had the positive correlation with the concentration.when concentration of PFP reached 5 mg/mL,scavenging rate of DPPH radical,hydroxyl radical and superoxide anion radical was 67%,59%and 63%,respectively.However,Antioxidant activity of PFP was week compared with VC.
